N-terminal sequencing of proteins and peptides by Edman degradation. Samples in solution or electroblotted to PVDF membrane.
.
.
.
Mass spectrometry. • Small molecule mass determination and/or structure confirmation (MS or MS/MS)
• Mass analysis (MS) of proteins or peptides in solution.
• Protein identification by nanospray liquid chromatography mass spectrometry
(LC-MS/MS) of protease-digested solution samples or SDS-PAGE bands.
• Post-translational modification determination (phosphorylation, methylation, etc) by LC/MS/MS of solution or in-gel protein digests.

SAMPLE REQUIREMENTS
.
.
.
Edman degradation. Minimum of 1 pmole. Samples electroblotted to PVDF should be visible by Coomassie Brilliant Blue staining. Call for procedure used to prepare samples.
.
.
.
Mass spectrometry. For solution samples, a minimum of 0.5 pmole, preferably in a small volume (~ 20ml). SDS-gel bands for in-gel digestion should be visible with Coomassie Brilliant Blue. Call for procedure used to prepare gel samples.
.
.
.
Bands on PVDF (Edman degradation) or from an SDS-gel (for in-gel digestion/mass spectral analysis) should be as concentrated as possible, i.e., maximum protein amount in as small an area as possible.
